Photonic Analytics Platform for Real-Time Monitoring of Stem Cell-Based In Vitro Models

As part of the PREPARE project »PremoCell,« the four Fraunhofer institutes – IPMS, IGB, IBMT, and IZI-BB – are collaborating to develop a modular biophotonic platform for a real-time analysis of drug effects in complex in vitro models such as stomach and liver organoids.

Fraunhofer IPMS is developing silicon nitride-based photonic sensor chips with the goal of continuously detecting selected biomarkers from complex biological models on a time-resolved basis.

To integrate the PIC sensors into preclinical testing models, the three other partner institutes are developing significant in vitro drug testing models of the liver and stomach at varying levels of complexity, with the aim of comprehensively addressing preclinical testing strategies through a high degree of flexibility. Fraunhofer IZI-BB is focusing on generating liver organoids from hiPS cells. These cells are to be further developed into a MASLD/MASH model by combining genetic risk alleles with defined environmental factors. Embedded in a microphysiological system with integrated real-time sensing, the models are intended to be used in the future for toxicity and efficacy testing of drug candidates.

Close collaboration within PremoCell is generating new approaches for preclinical research as well as for in vitro testing systems for material evaluation, such as within the framework of REACH.
